Human Leukocyte Antigen (HLA) part 22




Several techniques have been used to screen patient sera for alloreactive antibodies. The complement-dependent lymphotoxicity technique has two version.

One is a direct assay of patient antibodies exerting lymphotocytotoxic effect through complement-dependent  mechanism of cell membrane lysis.

The indirect technique utilizes an extra step, with goat or rabbit anti-human IgG antibody-mediated augmentation of complement-dependent lymphocytotoxicity.

This antiglobulin modification is more sensitive than the direct  lymphocytotoxicity technique, but is also more technically demanding. Use of enriched samples of B cells enables detection of HLA class II-specific antibodies.
